Effect of Mosapride Citrate and Combined Treatment with Ondansetron on Postoperative Ileus in Rats

|
Background/Aims: Postoperative ileus (POI) is a transient impairment of gastrointestinal motility after surgery. The mechanisms of POI are not completely understood, but it has been accepted that an activation of inhibitory neural reflex pathways was most important. Cisapride have been demonstrated to improve POI, but it was withdrawn from market due to cardiac toxicity. The purpose of this study was to investigate whether mosapride citrate (mosapride) and combined use of ondansetron could prevent POI.
Methods: Male Sprague-Dawley rats were used. Experimental groups were Control, POI only, POI+20 mg/kg mosapride, POI+mosapride with 0.8 mg/kg ondansetron, and POI+mosapride with 4 mg/kg ondansetron. POI was induced by massage of cecum under isoflurane anesthesia. Evans blue was fed after 1hour from operation. Gastric emptying (GE) and small intestinal transit (SIT) were measured after 30 min.
Results: GE and SIT significantly delayed in POI only group than control. Mosapride improved the delayed GE and SIT. The 0.8 mg ondansetron group showed more improved SIT than mosapride only group. The 4 mg ondansetron group showed more delayed GE and SIT than mosapride only group.
Conclusions: Mosapride have beneficial effect on POI and it was enhanced by low dose ondansetron, but interrupted by high dose ondansetron. (Kor J Neurogastroenterol Motil 2007;13:138-145)
